Aim of this work was to develop analytical methods for the peptide hormones IGF-1 and its analogues as well as the synthetic ACTH derivative Synacthen® for doping control purposes. Target matrices should be blood, plasma or serum. On the one hand, the great relevance of human IGF-1 is based on the fact that it can be used as a biomarker for the abuse of growth hormone. On the other hand, it can be directly applied in differents forms such as the recombinant drug, its analogues or in conjunction with gene doping. The spectrum of effects comprises mitogenesis, cell proliferation and differentiation as well as the inhibition of programmed cell death (apoptosis) in various tissues. Such effects are useful in disciplines where physical strength and/or fast regeneration are desired. Metabolic effects are e.g. improved utilisation of glucose, increased protein sysnthesis, reduced blood fat and diminished protein degradation. The analogues´ mode of action is qualitatively similar, but in parts more effective. The analytical method encompasses selective isolation by use of microparticle-based immunoaffinity extraction and liquid chromatographic separation followed by mass spectrometric detection. After administration, Synacthen® influences the physiology of stress. As an ACTH derivative, it stimulates the supply with endogenous corticosteroids such as hydrocortisone. Those provide anri-inflammatory and analgesic effects.This mode of stimulation is prohibited in controlled sports. The qualitative detection of Synacthen® used gel-based immunoaffinity extraction, solid phase extraction as well as high performance liquid chromatography coupled to electrospray mass spectrometry. Das Ziel dieser Arbeit war es, für die Peptidhormone IGF-1 und seine Analoga einerseits, sowie für das synthetische ACTH-Derivat Synacthen® andererseits analytische Verfahren zu entwickeln, die es ermöglichen, diese Substanzen in Blut-, Plasma- oder Serumproben zum Zwecke der Dopingkontrolle nachzuweisen. Humanes IGF-1 besitzt im Rahmen der Dopinganalytik eine große Bedeutung, da es zum einen als Marker für die verbotene Anwendung von Wachstumshormon dienen kann. Zum anderen kann es in Form des rekombinanten Präparates, seiner Analoga oder im Zuge des Gendopings als direktes Doping-Agens Verwendung finden. Es wirkt vor allem mitogen, stimuliert die Zellproliferation und -differenzierung und verzögert in manchen Geweben den programmierten Zelltod (Apoptose). Solche Wirkungen sind besonders in Bereichen des Sports von Nutzen, in denen Kraftkomponenten eine Rolle spielen und schnelle Regeneration erwünscht ist. Zu den metabolischen Effekten zählen v. a. verbesserte Glucoseutilisation, verringerte Blutfettwerte und verstärkte Proteinsynthese bzw. verminderter Proteinabbau. Die Wirkungen der Analoga LONG™R³IGF-1 und des(1-3)IGF-1 sind denen des hIGF-1 qualitativ gleichzusetzen, allerdings in z. T. erhöhter Wirksamkeit. Der Nachweis wurde durch eine selektive Isolierung auf Basis von Mikropartikel-gestützter Immunoaffinitätsextraktion und anschließende massenspektrometrische Detektion nach flüssigkeitschromatographischer Trennung erreicht. Synacthen® greift nach Applikation in die Stress-Physiologie des Körpers ein. Es stimuliert als ACTHDerivat direkt die Sezernierung endogener Corticosteroide wie z.B. Cortisol. Da Corticosteroide v.a. entzündungshemmende und schmerzstillende Wirkungen besitzen, ist ihre Stimulation durch Synacthen® im Bereich des Sports untersagt. Der qualitative Nachweis von Synacthen® gelang mittels Gel-basierter Immunoaffinitätsextraktion, Festphasenextraktion und Hochleistungsflüssigkeitschromatographie in Kopplung mit ESI-Massenspektrometrie. Die entwickelten Nachweisverfahren wurden umfassend validiert und anhand realer Proben erfolgreich getestet. Verf.-Referat
